新藤黄酸对宫颈癌HeLa细胞增殖、凋亡、迁移的影响及机制研究

【摘要】目的:研究新藤黄酸对宫颈癌He La细胞增殖、凋亡、迁移的影响及其机制。方法:不同浓度新藤黄酸 (0.00、0.63、1.25、2.50、5.00、10.00 mg/L) 作用于He La细胞72 h, MTT法检测细胞增殖;流式细胞术检测细胞周期;荧光显微镜和DNA ladder检测细胞凋亡;Transwell实验检测细胞迁移;RT-PCR、Western blot检测BCL-2、BAX、NF-κB及E-cadherin的mRNA和蛋白表达。结果:不同浓度新藤黄酸作用He La细胞72 h后, 细胞抑制率随药物浓度升高而升高;穿过Transwell小室的细胞数随药物浓度升高而减少;BCL-2、NF-κB mRNA及蛋白表达随新藤黄酸浓度升高而降低;BAX、E-cadherin mRNA及蛋白表达随新藤黄酸浓度的升高而升高。结论:新藤黄酸在一定范围内能以剂浓度依赖性抑制He La细胞增殖, 促进其凋亡, 并抑制其迁移。
