Chinese Pharmaceutical Journal is supervised by China Association for Science and Technology and sponsored by Chinese Pharmaceutical Association. Launched in 1953, the journal aims to reflect the progress of Chinese pharmaceutical and popularize pharmacy knowledge. This journal has a broad scope including biotechnology, traditional Chinese medicine and natural medicine, pharmacology, agentia, clinical pharmacy, pharmaceutical quality and inspection and pharmaceutical chemistry. Investigation of Key Factors Influencing the Extraction of Volatile Oil from Rhizoma Ligustici Chuanxiong by Steam Distillation

Objective To investigate the key process parameters affecting the extraction of volatile oil from Rhizoma Ligustici Chuanxiong by steam distillation. Methods Volatile oil and emulsified aromatic water were the products of the volatile oil extraction, and the quality of the volatile oil had direct influence on the formation of the emulsified aromatic water. Based on the analysis of effect of extracting temperature, drug-liquid ratio, crushing degree of medicinal material on the stability of the emulsified aromatic water and the yield of the volatile oil, the corresponding extraction process control model of the volatile oil was established to reveal the key process parameters during the volatile oil extraction and separation. Results With the rise in temperature, the relative viscosity, relative density, surface tension and contact angle of Rhizoma Ligustici Chuanxiong volatile oil decreased gradually, while the interfacial tension increased. The linear model of the Turbiscan stability index (TSI) of aromatic water versus process parameters was TSI = 0.877 + 0.230 × extracting temperature − 0.024 × particle size + 0.010 × drug-liquid ratio + 0.292 × condensing temperature + 0.776 × collecting temperature. The linear model of the TSI versus physical and chemical properties of the volatile oil was TSI = 0.877 − 0.170 × density + 0.098 × viscosity − 0.301 × surface tension + 0.695 × interfacial tension − 0.060 × contact angle. The linear model of the volatile oil yield versus process parameters was Yield = 5.065 + 0.258 × extracting temperature + 0.127 × particle size + 0.016 × drug–liquid ratio + 0.264 × condensing temperature + 0.264 × collecting temperature + 0.502 × TSI. The linear model of the yield versus physical and chemical properties of the volatile oil was Yield = 5.065 − 0.196 × density − 0.167 × viscosity − 0.201 × surface tension + 0.153 × interfacial tension − 0.065 × contact angle. Conclusion The TSI and the yield of the aromatic water of Rhizoma Ligustici Chuanxiong were directly proportional to the extracting temperature and particle size of medicinal materials. The yield of the volatile oil showed a significant positive correlation with the TSI value that reflected the yield of the volatile oil and was used as a specific index to evaluate the process parameters objectively.
